An overview on the protein folding problem will be presented. After the
introduction of the central dogma (DNA --> RNA --> Protein and Sequence -->
Structure --> Function), we will discuss the Anfinsen\'s thermodynamic
hypothesis which states that the unique native structure of a protein
corresponds to the global minimum of its free energy under the physiological
condition. Based on this, we formulate an ab initio method which is based
solely on global optimization of a potential energy function. Successful
applications of this methodology will be presented.
