생명체에는 유전정보의 전사 과정에서 RNA를 합성하는 RNA 중합효소가 있다. 본 연구진은 세균 및 세균바이러스 전사종결 연구에서 개발했던 단일분자 형광기술을 인간 RNAP3에 적용할 수 있도록 새롭게 고안하여, 인간 RNA 중합효소 III (RNAP3)의 전사종결을 연구했다. RNAP3는 tRNA, 5S rRNA, U6 snRNA 등 다양한 소형 비번역 RNA를 합성하는 중합효소로, 세포 내에서 높은 효율로 작동한다.
중합효소가 DNA 유전정보를 읽어 RNA를 합성하기 때문에 이 세 구성요소가 복합체를 이루어 상호작용하는데, 종결 시기에 이르면 복합체가 세 가지 경로 중 하나로 갈라진다는 것을 발견했다. 첫째, 종결자를 무시하고 지나치는 연속읽기(readthrough), 둘째, RNA와 DNA가 동시에 중합효소에서 떨어져 복합체 전체가 한꺼번에 해체되는 해체종결(decomposing termination), 셋째, RNA만 떼어지고 중합효소가 DNA에 남아 바로 재생 및 재개시가 가능한 재생종결(recycling termination)이다.
흥미롭게도, RNAP3는 세포 내에서 높은 재개시 효율로 작동하는 것으로 잘 알려져 있어 재생종결이 지배적일 것으로 예상되었지만, 실제로는 그 반대로 해체종결이 압도적으로 우세하다는 사실을 밝혔다. 또한 종결자 서열의 티민(T) 연속 길이가 길어질수록 해체종결보다 재생종결이 상대적으로 더 증가하며, 업스트림 RNA 헤어핀은 두 종결 경로 모두를 비슷하게 향상시킨다는 것을 규명했다. 이 결과들은 세균에서 인간에 이르기까지 이중 경로 종결이 진화적으로 보존되어 있음을 보여주면서도, 경로 선호도는 종 간에 역전될 수 있음을 시사한다. 또한 RPC10 서브유닛의 돌연변이가 두 경로에 서로 다른 영향을 미침을 확인하여, 두 경로의 기계론적 차이를 뒷받침했다.

Abstract: Eukaryotic transcription termination remains incompletely understood. Using single-molecule fluorescence assays, we examined intrinsic termination by human RNA polymerase III (RNAP3) and identified mechanistic trifurcation leading to readthrough and two termination pathways with distinctive dissociation kinetics. The dominant pathway, decomposing termination, involves (near-)simultaneous dissociation of RNA and DNA from RNAP3, resulting in complex decomposition. In contrast, recycling termination, where RNA is released while RNAP3 remains DNA-bound and one-dimensionally diffuses to enable facilitated recycling, accounts for 2–14% of transcription, diverging from its predominance in bacterial intrinsic termination. Confirming that robust termination requires a nontemplate-strand T-tract of four or more consecutive thymidines, we uncover that longer T-tracts favor recycling over decomposing termination, reflecting mechanistic distinction between the two pathways. While upstream RNA hairpins enhance termination efficiency, their impact decreases with distance from T-tracts, affecting both pathways similarly. Kinetically, transcript release takes consistently longer in decomposing than recycling termination, aligning with polymerase displacement versus RNA shearing. Furthermore, an RPC10 subunit mutation causes recurrent elongation arrest with differential effects on termination pathways, revealing its selective role in governing this branching process. These findings reinforce evolutionary conservation of termination-pathway duality and highlight diversion of pathway preference in human RNAP3 termination, establishing its fundamental mechanistic principles.
